I recently purchased a 67 Vette with the 427, 390HP engine. Yesterday I noticed some smoke coming off the left side of the engine back near the firewall. Using a mirror, I believe the smoke is coming out of a threaded hole in the block right next to where the oil sending unit is located. Should there be a plug there?

I would suspect you are having a little oil leakage from the valve cover gasket that is making it's way down to the exhaust manifold where it's burning off.

I think on the hydraulic lifter block there is a boss down there with a pilot dimple or hole that doesn't need a plug. The solid lifter blocks have threaded ports there for a remote oil cooler for racing. I'll see if I can find a thread with pictures.

Thanks for the photo. The hole that I was referring to is the 1/2" hole to the right and a little bit higher that the 3/4" hole

Pretty sure that is a blind hole. Shouldn't be anything leaking from it or smoking.

Should be a blind taped hole as you say so I didn't understand how smoke would come out. If it was drilled out believe there would be oil shooting out.
