Stop at Reds Eats in Wiscassett for their lobster roll !!
Boothbay Harbor is a nice place to stop and there shouldn't be much traffic in Oct.
LL Bean in Freeport is a great place to stop.

It has been many years since we were up that way but we always liked to stop in Camden for a few hours on the way to the Bar Harbor area. Quaint little coastal town with the harbor right behind the main street.
I never found Downtown Bar Harbor itself that interesting. Definitely geared to the tourist crowd.
If you want to eat or stay someplace classy and a little off the beaten path check out the Asticou Inn and Restaurant in Northeast Harbor. They probably don't do this anymore but my wife and I had the most incredible buffet there. La Grande Fete. They only did it on Thursday nights. Back then, and I'm talking a long time ago, you had to dress for dinner. Jacket and tie required. I didn't have two nickels to rub together but that night we dined at the table next to Burgess Merideth.

Camden is a great stopping place. Some nice B&B places there and, of course, great eats as well.
Bar Harbor Inn is the place to stay on MDI.
Don't be afraid to take a random left or right turn to see some cool places.
It's a really nice drive. Enjoy.

Cadillac Mountain for the sunrise is a great place to be for romance, or just a DJB...

Road construction into Ellsworth is a disaster. We haven't driven either Corvette to Bangor this year because of how bad the road is. If your car is factory ride height you may not have a problem but be careful none the less.
It is getting cooler at night and mornings (36 this morning) so bring some warmer clothes too. Leaves have already begun to turn so by the time you get here the colors should be in full swing.
Recommendations: For me I don't care to stay on the island because of the tourist traffic. I would prefer to travel up further into Downeast and see the country side. Lots more to see in Maine other than just MDI. Then again I live here so...

Check out the Seal Cove auto museum, not far from Bar Harbor.

Added some info to post #5 but then I thought you might not reread it.
Asticou Inn, Northeast Harbor. It doesn't look like it has changed much from when we were there.

On your way up or back near Camden/Rockport, the Owls Head Museum of Transportation. See link
http://owlshead.org/
You will be amazed.
Safe travels, and give us a road condition report if you can. I'd like to make it up there myself in a few weeks.
Thanks,

+1 for Cadillac mountain. Food wise I think the cheesesteak's Epi's in Bar Harbor are amazing. I go there for lunch and dinner. Jordan's is a good place for breakfast. I think they close around 2 or 3 though. Both food places are in the town of Bar Harbor.

Belated thanks all for the replies and info!
It was a great roadtrip to Portland, Bar Harbor & Acadia . Great meals and scenery throughout. The fall colors were in peak. Like driving thru a postcard. Only bad part of trip was some traffic at Ellworth (thx kg4fku) and major slow downs west bound Mass. and Conn.due to late pm sunglare on the return .
Miked- Stayed in Old Port. Every meal ++. Hit the shops and Lighthouses
DonVette- Your Grandsport is a twin to mine,
Bobs07- Red Eats lobster roll def. ++. Hit the LlL Bean (huge & 0pen 24 hrs?) and outlets in Freeport .
Jeffs82- No time to hit Boothsbay or Orchard beach. Maybe next trip along with Kennebunk ?
66db/c5firstimer- - No car shows or museums allowed as per the bride.
Scorch/Capthuff - Did Cadillac mountain twice for the sunrise. Great experience .Crowded with tourist and photogs. at that hour( Cap- not a good time for romance as you said. djb??? )
GreggE - Camden was beautiful. ++
