FOB 1 vs FOB 2
I have read a couple posts in which it is mentioned that the FOBs are different. One is #1 and the other is #2. How do you tell which is which and what is the difference?

Pull out the key and look down in the plastic portion attached to the key. There's a tiny 1 or 2 in there. It's hard to see but it's there

Actually, that is incorrect for 2016 key fobs. The center display on dash will show which Key Fob (1 or 2) is activated, when car is started.

Just started my 16 Vette in the Sport mode and I didn't see a 1 or 2 on the center display. Where on the display does the number appear? My 16 is a November 2015 build.

What trim is your car? It might be only on cars with the memory package. If I recall correctly it says something like "memory settings for fob 1 are being used" or something to that effect. It shows up on the center display.

The answer is those numbers have nothing to do with which FOB is which! Either FOB is FOB 1 depending on which one is programed as FOB 1!
I'll explain what the numbers mean:
It was ~6 months after I got my September 2013 built Z51 until a forum poster figured out what those key numbers mean. Surprised GM had not had the key vendor change the molds to A & B by now to avoid confusion.
A poster figured out they were die makes in a two cavity die. It's a random change on what you get!
Therefore:
25% get two 1s
25% get two 2s
50% get a 1 and a 2.
However this is the most confusion for 25% of the folks who look, half of those who get a 1 & 2 have the first programed FOB be different than the key number!! For those folks, just swap keys!!! It would be foolish for GM to put numbers in a key that can be swapped- think about it! In fact 25% of folks who had a 1 & 2 and by chance had them programed correctly were a PIA! Kept telling others we didn't know what we were talking about. Some brought keys back to the dealer - who had no idea!!
Reminds me of the folks who told me I didn't know how to have my 2014 hatch latch, the seal would break-in or I was just not strong enough! When I sold my 2014 a few months ago it still would not latch unless the door was open! Those of use that had the issue were vindicated in 2016 when GM reverted back to the motorized pull down I had on my C6 and had to swallow all the "hype" about the magic memory wire operated vent! Never did tell us why some cars were more air tight than others! Happiness was a tight Vette!

If you don't have memory recall would it make a difference?
My fobs 2016 don't have numbers in them.
I only got one fob when I bought the car.
Got a new one and did the long programming so I would have #1 and #2
fobs programmed instead of #1 and #3 if using fob detected programming.

I have 2 #2's as well, but I keep the 2nd one in the house safe

I did a trial and error. I have 3LT with memory seats. I programmed seat memory #1 in an extreme position and activated automatic seat adjustment, returned the seat to "normal position", got out of the car, locked it and back in again to check if it moved during entry and exit after starting up. When it didn't I knew I had the wrong FOB. I then marked the FOBs so that my wife could have one with her settings on it.

This is what I was told during museum delivery: The FOBs aren't (I think) distinguished by the little number. There's a big colored circle at the end of the FOB. For me at least that circle is yellow on one FOB, green on the other. There shouldn't be any need to mark or guess which FOB is which.
The fun came driving home 1500 miles with one FOB in my pocket, the other in a duffel in the hatch area. The computer was so confused. It would chime and honk seemingly randomly every time I got out of the car to warn me there was a FOB inside the car. Memory recall would randomly pick up one of the two FOBs and often just fail to recall. All that got sorted when I got home and put the second FOB inside the house.

I have a 1LT (no memory anything) and have a #1 and #2 in the FOBs. Been using the #1 all along and didn't know it, but it doesn't matter anyway.

Bought a silicon FOB cover (Amazon) that keeps the key attached to the FOB. Transferred it from my 2014 to Grand Sport FOB. It comes in colors so you can let your wife pick!
Note: My Grand Sport key does not come out as easy as my 2014 but suggest you check it see how the latch holds. I recall my C6 FOB key was very difficult to remove. It obviously depend on the latch.
