What about the opening on the opposite side of the manifold....closer to the side of the manifold that is closest to the windshield?

if you are talking the rear of the manifold, there is no coolant passage there if i am reading your post correctly.....the passage is in the front, so anywhere there is ok...or cylinder head...........if you are talking about tapping the rear passage in the head that is blocked by the manifold, i dont think you have enough meat on most manifolds??

What exactly is an engine thermometer? Does it measure water temp? Where on the manifold is it located?

it is permanently mounted in the intake next to the thermostat housing and takes all the guess work out of correct engine temperature...the dial is calibrated from 50-300 degrees........

Now, in my big block the hole next to the thermostat housing is the heater hose port. My temperature sender is in the left head same as the small block.

The temp probe located at the thermostat is the best location, because it is the temp of the coolant (the warmest it will get) just before it goes to the rad.

i am using the transdapt thermostat housing that has the threaded hole in it to mount my electric fan temp probe.

when i compared it to the temp sender to the dash in the intake right to the left of it. there was only a 10 degree difference in temp.
