Sounds like a already ready to go car... you might want to double check the electric side of it... and the water hoses... with 5 hours drive, it s gonna be under volts and pressure quiet a bit.
Check all fluid levels (oil, master cylinder, windshield washers, coolant). Check all lights, wiper blades, tire pressures (including spare).
And if things don't go as planned: cell phone charge, AAA card.
I do not consider a 5.5 hour drive to be an unusually stressful condition. Normal maintenance and inspection should be sufficient to prevent most umpleasant surprises.

Just went 12 hours in my 02Z. Checked the oil and tires, filled her up with gas and hit the road.
In most ways, a 5.5 hour highway drive is less stress on a car than a 40 minute stop and go on the way to work.
If it's a car you don't drive much there are the normal things to check and go over but if you drive the car 3 - 4 times a week a 5 hour drive should be a piece of cake If you have done up grades get some miles on it before you go to make sure its OK
Geez you guys forgot the most important part... cigars. 5.5 hours... that's good for 2-3 cigars at least! Oh and don't forget your iPod... and some toilet paper... you never know!
I always have a little travel box in the back. Couple of quarts of oil, tranny fluid (auto), windex, paper towels, a few tools, and the like. Pair of cheap cloth gloves. Small terry towel. Bottle of car wax if I get the urge.

Shoot I drove my 92 500 miles to the track on Friday and 500 miles back on Monday, along with 200 track miles.
Yep I've recently changed the oil - it was time.
Bleed the brakes to get out the old fluid from the last two events.
Now I'm going to a track 4 hours away on Sunday and I'll drive back Monday night.
Won't do anything but bleed the brakes a little and check the pads.
The thing that comes to mind is to make sure your repairs have been in place for a couple of weeks...nothing like a coolant leak or something to break you down in the middle of nowhere.
