I have read/seen 2 different methods. 1 is to tighten it down then tighten the poly lock. The 2nd was to tighten, then tighten the poly lock, then give one more 1/8 turn on the whole body after tightening the poly lock.
Get your lash set, hold the wrench and tighten the set screw. Then Bump the wrench with your hand. This tightens the whole nut and set screw and seats it. You can put a dab of locktite on set screw if you want, but not needed.
I also run a paint mark on each nut and set screw. This way if I have valve covers off I can inspect if anything loosened up.
