I have heard both sides of this argument.
The way I see it is if GM wanted you to only use drive "D", they would not have included first "1", second "2", and third "3" on the transmission selector.
These cars are designed and tested, and tested again. I seriously doubt that the GM engineers would allow you to shift the A4 if it caused premature ware.
I think that alot of people buy into the myth that the A4 is fragile. From my personal experience, it is quite the contrary. Think of it this way, how may posts do you see about the A4 blowing up? Not many. And most people have mods around here.
Dont get me wrong, the A4 can blow up. Either though a defect in the transmission or throwing too much power at stock internals.
I shifted the AT tranny all the time in my previously owned Mustang. Now GM A4 may be a miracle tranny. But my Mustang's tranny go messed up with all this shifting within 14-17K miles or so. I got it rebuilt, and was told never to do it again. I also did some research on Hot Rod, and other sports car magazines. Don't know exactly which magazine I read, but it was an article CLEARLY for shifting AT trannys. It was highly NOT recommended. SO now, I don't shift my A4 on the Vette, but once in a while I do do it. Thats the reason why I'm thinking of changing AT oil/filter now to always keep it running in good shape. I'm not an expert, but my 2 cents...and what I've learnt
I have shifted my '93 and '97 Sevilles and my '99 vette ALL the time, more often than not, and not ONE single problem in over 100k miles of combined driving.
I shift my A4 all the time. 16K miles, so far no problems.
My 95 Impala SS (daily driver) has been shifted since I got
it new 10 years ago. Never been a problem for 115K miles.
Upshift and downshift mine all the time and never had a problem. Somtimes I find myself sitting at a light too long and pop it into "N" to relax my right foot from the brake for a while then when the light changes, I'm off shifting her again as needed.
There are many more reasons but I do it mostly to save the brakes by allowing the trans to slow you down, quicker response when cornering or to hold RPM when needed.

My A4 lost 3rd and 4th gears at 40,000 miles. Without abusive driving. I never mentioned it before and have heard of many others breaking.
To keep the motor at its max power band, just in case you may need to smoke another car from a roll. Of course I would never need to use this method because racing is illegal.
