I have a house there on Isle of Palms, about 15 minutes outside of Charleston. Absolutely the best place I have been to in these entire 50 states.
What to do? Wow....
Go to the beach. Take a tour of Fort Sumter. Take a dinner cruise in the harbor. Go on a carriage ride through the historical district of downtown. Walk around downtown. Go to the battery. Take a tour of one of the many plantations (Magnolia, Boone, etc.) Eat tons of seafood (tons of great restaurants). Go to the market. Drive to one of the many islands surrounding Charleston (James, Daniel, Palms, etc.). Shop downtown. Golf. Go to one of the many pubs. I'm sure I'm forgetting a ton......
Not sure what you are thinking in terms of where to stay. But Wild Dunes has MANY options. A little pricey, but if you want to 'do it right', you can't stay at a better place really. Right on the beach, 10 mins to downtown. Surrounded by 100's of restaurants (within 15 mins).
You really have to go 2-3 times to get the full experience. Feel free to PM me for more details!

The Holiday Inn near downtown is nice, not your traditional Holiday Inn. I don't remember the address, but it's the one near the Aquarium. You will be within walking distance of the market, pubs, the aquarium, and museums. Have fun.
Go to Patriot's Point and see the USS Yorktown. WWII aircraft carrier nicknamed "The Fighting Lady" also visit the museum that houses the CS Hunley. It is one of the first submarines to be used in naval combat during the Civil War. (act of Northern Agression) The CS Hunley sunk at least twice killing both crews and is actually hand powered! The CS Hunley was recently recovered thanks in part to the famous author Clive Cussler. You can read about the CS Hunley in his book: "Sea Hunters" where he actually explores several sunken boats and some of the Civil War monitors as well. "CS" stands for Confederate Submarine according to Cussler's book.
