I'm replacing the Optima Battery in my 2000 convertible with the factory original AC Delco. When putting in the new battery, will the onboard computer have to be reset? If so, does this have to be done by a Chevy dealership?

Depending on how long you have the battery disconnected you may or may not lose your radio station station presets or your radio may go into lock mode. Just make sure you have your radio security lock code number beforehand, just in case. Resetting your station presets/clock and unlocking the radio only takes a few minutes and isn't hard to do.
As far as everything else, your PCM takes care of it with a little driving time.

Thanks guys for all of your responses. Hooked up the new Delco last night and everything is ok. Didn't lose a thing. As far as going with the Delco, I just want to take it back to being a Vette with original parts.
