My wife and I did a 3,300 mile trip in Sept from NM and on through the mid-west and back. Ours has approx. 95k miles and ran flawlessly.
In addition to the basics and if you have a 6spd. I'd recommend following the popular post on changing clutch fluid before you go. If you're belt isn't too bad buy a spare one and throw it in the compartment out back with a set of tools.
Also - advise you purchase a radar detector cause of course your gonna speed at some point on those open roads w/o anyone else around. This small investment saved us thousands.
We don't have run flats so made sure to have a can of fixaflat. I also had some compact co2 tire inflators (from my dirtbike days) just in case. Be prepared to scrape the bottom of your car here and there, take on a couple road chips on the chin and watch out for the semi-truck tire debris - seems is too common out there and wondering why a better standard isn't placed on these tires.
Driving the vette was a blast for us. Smooth, comfortable and exciting to drive . We debated on whether to take it vs. the "68 Camaro and glad we went with the vette on this trip.
Next year we'll be taking the '68 Camaro through Utah & AZ.
I gotta say I'm jealous, work has been a drag lately and a nice long road trip w/the Vette sounds real good and it's a great time of year to do it! If your car has been running good then get an oil change, a GPS and GO BABY GO!
Jimman drives his 2007 Z06 all around the country. He has around 120k miles on it already and the only thing he had to replace was an oil pressure sending unit at 98k miles.
Change oil/filter if it has not been done recently
At 50k miles, front brakes may be due, check for pad thickness
Has the coolant been changed? They say every two years, but at your miles and time, i'd change it.
Check the inside of the belt for any small cracks in the rubber, if it has not been replaced before, i would replace that.
Squeeze the radiator hoses when engine is cool. If its spongy or you feel any cracking of the hose, replace all the hoses.
Tires, you may have good tread, but check the age of the tires. by looking at the sidewall, you can normally tell the age (year made) by looking at the dot or serial number on the sidewall. Tires over 5 years old can come apart especially at highway speeds and hot temperatures. I had a 6 year old tire with great tread come apart on me this past june enroute to the power tour back east. I made it from so. cal only to vegas when the tire came apart.
I always carry a tire plug kit, small compressor, small hand tools, gloves, water (for me or the car), cell, charger for phone, any meds, check wiper blades (they always get forgotten, at least in ca), age of the battery? batteries in key fob? extra key/fob.

My wife and I with another couple (C6) did a 5,200 mile trip this Summer. I did an oil change before (and after the trip), checked the Tire pressure and went. Get off the freeways as much as possible, better scenery and more twisties.
I left Virginia a couple of years ago with 37,000 miles and came back from the west coast a month and 8,500 miles later. Had the car tuned and checked hoses/belt before leaving. Drove Rt. 66 then up Cali, Oregon and east again. No problems at all. I'd join AAA just in case (we did, but didn't need them).
I did a 2,800 mile trip solo in September. I needed new tires, so I did that and an alignment but after checking the car over, I just drove it.
Only other thing I did was to add a Double D mods MP3 kit to my factory Bose stereo so that I could play the MP3 player and Pandora radio app on my Andriod phone. It was well worth it.
