I would say YES.
When I replaced the coolant, I had a heck of a time getting the air out. It took several "burps" to get a consistant temp reading.
Do a search and follow the instructions. It is not that difficult.

Just the jug? Did you drain the coolant prior to? If you did NOT drain the coolant and removed and replaced the jug, then I would say NO. If you did drain the coolant, YES.
Here is a tip for you C5 guys trying to bleed the cooling system. Fill the engine with coolant with the upper radiator hose
