Depends on tuning, final horsepower numbers and your driving style. Plenty of guys on here with those mods who have been running them without issues for a long time.

Check with the shop/vendor you plan to use. They should have a good idea if any changes are going to be needed. But it does appear that a number of owners have done the upgrades you mention on a stock bottom end and are doing OK.
with just the exahsut/super and cam, you'll be very close to 600, if not more (depending on how much boost you go with)... my ls1 is at 530 with just exhaust/cam/aa super.
the cam I used is mild and we chose it specifically to fatten the toque curve at the bottom end. My guess is if you also do heads, you'll be *well* into the 600hp range.
Don't listen to the naysayers -- especailly if you're not tracking it, the ls6 can handle 600hp all day long. What kind of trans? I assume manual, same, zero issues.
