LS6s have stronger blocks, but otherwise, performance-wise, most things are the same. The heads, cam and intake manifold are the main differences. The fuel injectors are slightly better too.
the LS6 has the "little things" that make a relativly little HP difference (for the price). The first generation LS1 engines had 345 HP or the later model (01) which got 350 HP. The LS6 introduced in the Z06 for '01 pumped out 385HP. (source: Corvette Chronicles) I dont' think it was until '02 that the LS6 was beefed up to get that precious 405HP.
They have head and cam kits to go to an LS6, but don't waste your money.
Your time and money is better spent buying an aftermarket cam and heads if you are going to do the work.
You will also get a lot more power out of it.
You would need heads, cam and an LS6 intake, gaskets, etc.
Let me know if you need any specifics.

Add to that. LS6 oil pump, timing chain, under drive pulley while you have the stock pulley off.
But as mentioned go with after market heads and cam for better HP gain. I've been there and done that. Waste of money on stock LS6 heads and cam. My $.02
