The 4L65E is a FOUR speed transmission from a 2005 C6.
I have NO experience with using this transmission in place of the 4L60E.
If you want my "Guess" I would think it is possible with minor modifications.
Six-speed auto from a 2006 and up - no way. Maybe dropping the entire C6 drive train in along with the computer, etc. would work. But why - with all the pain of converting EVERYTHING.
The first year C6 (2005) used a FOUR speed automatic. 4L65E (4 is for FOUR speed)

P.S. The service manual refers to the 2005 auto trans as a 4L60E/4L65E
So what exactly would be the point of swapping 1-4 speed auto for another here then? I assumed the original poster meant the 6 speed if he was going through the hassle (probably wants the paddle shifter like the new autos).
4- 4 forward gears
L-means trans is installed lengthwise IOW RWD (T is for FWD)
60-scale for it's torque capacity. 80 is stronger than a 60, etc
E- electronic control
This means a 4L60E is slightly weaker than a 4L65E. From what I understand, it's mostly the same transmission with a few minor adjustments and improvements to make it stronger than the 4L60-E. I've heard of folks upgrading their 60's with 65 parts. I would say it would work fine, but do your research. There could be mounting differences or connector differences or incompatabilities too since it's electronic.
I would say call a tranny shop that deals with lots of C5's and C6's. They should be able to steer you in the right direction
as other people have said, i read that the 2005 c6 tranny is a slightly better design. i need to work on mine as i have lost gears due to wear in the valve body. so i thought why not go a year newer if it was straight forward.

I hear ya - No advantage that I know of, but he asked about a 4L65E, so I thought maybe he had access to a 2005 Corvette transmission that he wanted to use as a swap.
