I don't believe the c5 speedometer calibration doesn't change with a gear change as the speed sensor gets it's signal off a toothed wheel of the ring gear.

If your going to the trouble to change the axle ratio why just go to the 3.15?
You'll barely notice the difference, step up to the 3.42 and then you'll have something.
BTW, what will need to to be changed is the shift points and that can be done by anyone with tuning software.
Tech 2's are very limited to what they can do for a permanent change.
The reason I'm asking is the car has already been dyno tuned by a professional tuner but he moved out of state and the only mod this time is the differential. If the Tech II can change the broadcast code and thus the programming of that one item it would be ideal...no retune or other hassles.
Oh, and I'm going with 3.15:1 gears because I'm getting the differential in trade for something.

You don't need a tech2 to change anything. You need to get your tranny re-tuned to adjust your shift tables.

My understanding is the factory C5's came with both 2.73 and 3.15 rear differential options and thus have BOTH shift tables programmed as OEM shift tables. The difference is in the PCM programming broadcast code for either one. I was told since it is ALREADY in there for these two optioned differential ratios ONLY that the Tech II could program the change.
Now that I've explained in depth what I'm looking for...can anyone tell me if it is possible? Thanks again all who have offered assistance so far...

Awesome...you are the man. Without getting too much into it a dealer "friend" says because it's only going to the 3.15 gears he could reprogram it at the dealer after hours with the Tech II by simply reprogramming an RPO/broadcast code from G97 to G92. Just double-checking if it was even possible as I had always heard differently.
The Tech II will only program to your vin #.
So unless your car came with a 3.15 axle no it won't make the change, it can be done with some handheld programers for 99 and up MY's.
Otherwise you need tuning software to change the PCM.
