I'm about to be putting headers on a C5 but the only thing I've ever done before were F-Bodys. With those cars you can completely remove the rear 02s and just use the front sensors but you need a tuner to delete. With a Vette is it the same thing?

Yes, and it's no coincidence.......it's the same engine.
Yes you will have to delete all the rear 02 sensing codes--
LT headers also open up a whole new can of worms !! Because on a C5 the front 02 bungs are much further back then they were on the stock manifolds-sometimes as much as 3'----You often get slow or lazy 02 codes because of the further back location-Many people to correct this will install the REAR 02's onto the FRONT location as the rear ones are more sensitive and heat up better---
Another byproduct of the further back bungs is that your P/T or closed loop fueling
becomes richer than the Stoich 14.7--It settles around 14.0-14.4-(That is why they are illegal in most states)-- Tuning will help get it closer back to 14.7 but it never gets it--There is NO 100% fix--(other than installing wideband 02's on the front) but most just settle with what they get---Doesn't hurt anything engine wise--just produces more hydrocarbons and uses more fuel
