Speed costs money, how much do you want to spend? :D
Seriously, it's all a matter of what your budget is. In order of what I'd look at:
intake (zip tie mod, Blackwing, etc)
exhaust (headers, not mufflers. mufflers will improve sound but won't gain much HP)
cam

I'd either go with:
1. Good intake, headers, & tune
2. Cam & tune
Personally, with a Z06, I'd probably go for the cam & tune. I'm actually thinking about this, and I've seen plenty of guys with no other mods that have picked up close to 50HP on Z06s with just a cam change.
