Its probably full of sand and grit and stuck. Theres a DIY on how to take it out and apart to clean it. I have to do mine about once a year. Never mind. I missed the part about not working from the dash.

Check to see if you have +12v at the release motor when the button on the dash is pressed. If you do, you may have a bad release motor. If not, you may have an open circuit some where.
My C6 trunk release won't open....using the button in the car or on my fob....only if I put the key in under by the plate??
Any ideas?

I looked in your profile and it appears that you have a convertible. The convertibles and early Z06's have a manual latch. The other coupes and Z06's built after about 2010 have an auto pull down latch. I took these pictures when I was installing an auto pull down latch into my 2006 Z06.
In a convertible or an early Z06, you'll see panels exactly like in the pictures.
What you're interested in checking is the blue 15amp fuse in the sub-block under the cover to the right of the main fuse block that I have a circle around.
If that fuse is blown then you'll have exactly the problem you're experiencing.
If the button above the license plate is dirty and not working, usually the button in the cockpit will still work - unless the sticky button blew the fuse, then nothing but the key will open the trunk.
