What is the best method for placing the C6 into accessory mode for an extended period of time? I have two batteries so running the battery dead is not an issue. However, would like to play the radio without having to reset the accessory mode every ten minutes.
Hold the bottom of the button a bit longer to turn the ignition "on" instead of accessory mode if you need to use it for extended periods of time. Battery drain will be much worse because it turns everything on instead of just the creature comfort stuff. Turn off the headlights for sure.

Just plug a battery maintainer in while in accessory mode.

It's not a good idea to have the ignition on without the engine running for extended periods.
Doing this will cause premature failure of the ignition system.
The ACC mode times out and this behavior can't be changed. I also agree that turning on the ignition for long periods of time is a solution but a bad idea. I have never tried it but the ignition may also time out after a while when the computer detects that it is on but the engine is not running. Who knows how long that timeout may be, if it exists at all.

The safest thing for your vette would be to rewire the power to the radio. If you put in two batteries and all the associated wiring, that should be easy for you to do
