2006 c6 Driver door all of the sudden will not open when button is pressed. Window indexes down a bit but door will not open. Press button again and window goes back up and the process repeats. I need to use the emergency release to get out. No other issues. Any ideas, suggestions or help, greatly appreciated.

It could be as simple as the finger pad dirty with grime.
Take it apart and clean it. If that is not the solution, you may need a new door latch assembly.

Sorry that I misunderstood the issue. It could well be the interior switch wiring or malfunction.
From the Service manual
Door Latch Inoperative from Interior Door Handle Switch
1. Ignition OFF, disconnect the harness connector at the inoperative interior door handle switch.
2. Test for less than 1.0 ohm of resistance between the ground circuit terminal A and ground.  If greater than the specified range, test the ground circuit for an open/high resistance.
3. Ignition ON, verify the appropriate door control module scan tool interior door handle switch parameter is INACTIVE.  If not the specified value, test the signal circuit terminal C for a short to ground. If the circuit tests normal, replace the appropriate door control module.
4. Verify the appropriate RCDLR scan tool interior door handle switch parameter is INACTIVE.  If not the specified value, test the signal circuit terminal B for a short to ground. If the circuit tests normal, replace the RCDLR.
5. Install a 3-A fused jumper wire between the signal circuit terminal C and ground terminal A. Verify the appropriate door control module scan tool interior door handle switch parameter is ACTIVE.  If not the specified value, test the signal circuit terminal C for a short to voltage or an open/high resistance. If the circuit tests normal, replace the appropriate door control module.
6. Install a 3-A fused jumper wire between the signal circuit terminal B and ground terminal A. Verify the RCDLR scan tool interior door handle switch parameter is ACTIVE.
If not the specified value, test the signal circuit terminal B for a short to voltage or an open/high resistance. If the circuit tests normal, replace the RCDLR.
7. If all circuits test normal, test or replace the appropriate interior door handle switch.

I have the same problem on the passenger side. Opens from the outside but only indexes from the inside, my car is in storage right now but if you figure out what the issue is please post your solution! I tried replacing the interior door button, but no effect. But I do have a spare interior button now

 

I replaced the battery in the FOB but still door only partially released from outside. My only thought was battery might have been a little low from sitting. I put my maintainer on the battery and seems to be OK. Just curious if related to flash after catalytic converter recall that I had done by dealer??

Unfortunately I'm on the opposite side of the planet until next summer, so I can't troubleshoot. I did find an old thread though where someone mentioned that the issue is actually caused by the exterior pad going bad and somehow it effects the operation of the interior button.

 

My guess is that it's unrelated to the flash, but it will get worse. Mine started by doing it intermittently once in a while but would open on the 2nd try, and eventually got more frequent until it just stopped opening completely and only indexes no matter how many times you try.

 

The easiest thing to do first is remove the passenger side button and plug it into the driver side door and see if that works. If the problem still occurs you know its not the switch. If you have the same issues its time to check the latch release function. Do not insert passenger switch into driver side opening...just let it dangle. You can also test the driver side release by plugging it into the passenger side door. Again, just plug them in and let them dangle. Do both at the same time.
