Improvement in what horsepower? No. Other factors will depend on your mods and how they tuned it on the dyno. If you have some edgy mods working with it on the street can help drivability. But, it not necessarily going to give you any more power.
how was it dyno tuned ? lots of various loads, runs, etc. over a number of days, etc..???
I had my car to a dyno tuner once. He strapped it on did 2 WOT pulls and said ok, here ya go. It was just a header cam car at the time but I would have liked to see a little more time spent on it. My drivability wasn't that great and his "canned" initial tune for a header cam car just didn't cut it.

they did over 15 pulls and 5 hours with mine but that was in the early days of the edit programs for the C6.
My car was dyno tuned April 06, and the tuner said he didn't have everything on hand to properly tune the 06's and to have it retuned possibly in Dec 06....Jan 07. I had to drill a 1/8" hole in the throttle body plate for it to idle properly. Currently, I have no problems with drivability.
Over 90 pulls on my Mustang, with 3 hours of street time. Blower car that I left at the shop so the guy could use it for testing. Of course a daily business can't spend that kind of time on a car... but there should be some kind of comprimise. He used my car to find base tunes but ever car is different and still required a few hours each. When I got it back it ran BETTER than stock. It was amazing. On the street tune (detuned) it made 620/599

Got a hole in my 05 also. The throttle cracker tables are not there like they are on the C5. The 06 cars are even more locked up. No VE table there yet I think.

PM your phone # and a time to call. I need to discuss your comment.
My car has been dyno tuned at 462.5 RWHP. Will there be an improvement if I get a tuner to do a tune while the car is being driven?
My personal opinion is street tuning is superior to tuning on the dyno. When driving on the road/track more air is flowing. IE -Varam or a CAI, the ram effect will increase airflow to a point that it can't be recreated on the dyno. The load is also different on the street... I think the dyno is a good tool tool for checking A/F ratio's and performance gains... If you do have it tuned on the dyno, I would reccomend usning a logger on the street to check knock retard.
as for which is better I think you'll see more & more tuners doing street tunes in addition to work on the dyno because of the fact that you really cant simulate airflow & load on the dyno as well as you can on the street. I already know of a few in my area that are doing both.
