You can, as long as the sub doesn't interfere with the compartment cover on excursion. You can build a cover with a integrated speaker grill or you can loosen the factory cover when playing your music to allow more air movement for the sub and to maximize the distance for excursion of the sub.
I'm thinking about buying a 2nd factory cover then cutting a hole in it and adding steel mesh.
I would like to keep this pretty stealth looking while keeping the functionality of the hatch area but I also don't want to be interfering with the subs excersion and air flow.

This is something I made and it serves the purpose. Pretty much the same cover as what JL uses, except I use a heavier gauge steel mesh with more perforated holes to allow the best airflow.

I used Fiberboard and I believe it is 1/4". I can measure it and make sure this evening. It's pretty strong! I just popped out the original latches in a stock compartment cover and used them in this. I think you can purchase them through Fred Beans or your local chevy dealership, but am not positive.
I have a relatively thin sheet of aluminum that my fabricator friend used to make up a cover for me. I raised it about 1.5" over the factory floor height to make sure there would not be any issues with excursion on my stealth box (no chance it would get there but wth). The shape is a flat at the factory location at the floor level and a raised square over the enclosure. I will find out how thick a sheet he used and we made a tightly spaced circular pattern over the top of the sub to allow air flow, but I believe I am going to do something similar to what NV did on his cover to improve the airflow a bit. It does hurt sub performance a little just so you know.
