I'm sure others here who are better informed will chip in but I would have thought that a 600cfm carb would be on the small side for a 383 stroker. I have a 350 putting out 430hp and have an 800cfm carb on it.
Carb is going to be too small. You will most likely need a 750. As far a trans if the th350 is in good shape it should hold up. A th350 will hold up to your engine it will just depend how much time yours has left.

I ran a big block 427 pushing 500 horses with a Turbo 350 and had no problems. Just make sure it's in good shape. I have a 383 stroker in my 61 with a Speed Demon 650 double pump. It feels a little under carbeurated. I would go with a 750. You will enjoy that stroker. They are stump pullers.
