Check to see if gas is squirting into the carb when you pump the accelerator. To do this remove the air cleaner. Look inside the carb while rotating the accelerator mechanism on the carb. The accelerator pump should squirt gasoline into the carb. If you determine it has gasoline in carb; it could be a timing issue.
I had the same problem with my 1980 when I replaced the motor with a 383 (de-stroked 400 in my case). It turned out my fuel pump had failed. Perhaps removing it after 28 years did it in. I don't know, but I bought a new fuel pump and it ran.

it was a custom built engine, has already been run on a stand, I managed to get hold of my engine builder, I didnt prime the carb, all the fuel in the fuel line would be gone by now, car sat in garage without engine,for about 5 months, so I will try prime the carb 1st, then once its started fuel pump should start working at full stream.

Dave, did the engine builder run the engine on a test stand with an electric fuel pump (but not your mechanical pump) if so - was the pump push rod installed?
You can check by removing the access bolt just below the pump housing!

I asked him that question, yeah he run the motor with the fuel pump thats on it just now, I will try primeing it tomorrow.
update - put some fuel in the carb bowls, engine started, pump started pumping plenty of fuel, job done, but I have found another problem, a small oil leak!, I will start a new thread for that one

apart from a small oil leak, (which I hope is just the oil pan seal, and mot the rear main seal, all new seals!)
the engine is running great, revs crazy fast for a push rod engine, great sound, really pleased with it, not had it out for good spin yet, still trying to put everything back together!
