I'm in the process of restoring the smog pump on my 1970 LT1 mostly for cosmetic purposes. ---- My question is: should I remove the side cover and clean and lubricate the rotating components while I have the pump off the engine?

I never thought of your question before so I looked in the Chassis Service Manual to see if there was any info about maintaining the pump properly ... not a thing!
So I don't know.

If I remember correctly, the cover is aligned with one or two dowel pins. Remove the bolts, slide the cover off of the pins. There is a big bearing that is either part of the cover or part of the cylinder inside the pump. The bearing could probably require some grease.
The bearing on the front of the pump is sealed so you can't grease it. There are carbon shoes that rub on the vanes inside the pump. They don't require lubrication either.
