Is there any way to modify a Mark 4 big block to use Mark 6 factory hydraulic roller lifters?

If you want to run a roller in a MK 4 go to competition cams web site and check out what they offer. Arent the factory lifters to long for the lifter bore in the block amoung other possbile issues

I dont think you'll find any easy way to do that conversion.
Fortunately all the cam companies sell link bar type hydraulic rollers for Mark IV applications.
