A lifter will usay not be a knock but a clanking noise for lack of better terms and noticable right under the valve covers. If you have a rod or crank noise you can usaly locate the cyl by taking the spark away that way it is taking the load off that cyl. you can always use a sephoscope (spelled wrong) to help locate. Spun rods are way more common then mains.
