I am trying to finished off my carbed 383 build in my 88. I am running an intake manifold that locates a substantial amount of the coolant above the radiator cap.
I have decided to use a Moroso radiator hose filler (63745) install in the upper radiator hose to fix the problem.
I am assuming that I need to relocate my expansion tank hose to the new fill point. If this is wrong, correct me.
My question is what do I do with the old filler cap and expansion hose outlet? Cap the outlet and put a high pressure radiator cap in the factory location?

Why not just tee into the existing overflow hose and leave the standard cap and overflow hose in place.
It will be fine tee'ing into it. You have a pressurized source going to a non-pressurized location, so it will always flow toward the tank, even if both caps open.
I'm not sure why you need the hose filler. The TPI manifold held water above the filler cap too. The upper hose is the water return from the motor to the radiator. L98s are essentially self burping. I carbed my 84 for years and never had a heating problem.

This all started from what appears to maybe have been a faulty cap that has since been replaced...

With the reverse flow Lt1 motors it is important. Your L98 should have no problems.
