I'm sure there is a thread on this somewhere but I may as well jump in here and ask my question. Do you break the brake line to do the Rustoleum paint job, meaning, does the caliper have to be completely removed from the vehicle?

You can, but it is not necessary.
I didn't and they turned out great. Just tape off what you dont want to get paint on. Take your time as this is the most important part of the whole process.
And to remove the paint from the lettering, someone suggested an exacto knife. I am sure that would work, but I simply used some "Goof Off" on a rag and removed the paint that way.

Just buy the paint and do it yourself. I went with Folia Tec paint and total cost was about $50. Sure beats paying better than $300 for the same calipers you have on the car now, only red.

A quick question on the painting. Everyone talks about taking the paint off the lettering with either an exacto knife, sandpaper, or paint remover of some sort to get back to the "metal look".
I have a pewter vert and am planning on painting mine shortly (planning on black). I realize that the paint is a high temperature paint, but was wondering if I could use body paint for the letters. What would happen if this paint was not the same high-temp stuff? I assume that it will eventually flake off, but I am looking at using it more for show purposes. Would it potentially damage the wheels (magnesium)?
Also, someone has mentioned applying some sort of a clear coat. Anyone have experience with this?
As stated earlier, I painted mine with Rustoleum using a foam brush. Did the calipers red and then using the edge of a foam brush by lightly passing it over the letters, I did those in black. It's been 2 years and none of the paint has flaked off.
Clean 'em real good with brake cleaner and a rag. You wont need a big can of Rustoleum, buy the small one. I went to the hobby store and bought a camel hair type brush.(more paint control) For the letters I used a Testors silver paint pen..... worked great! P.S. It took 2 coats of paint per caliper to get good coverage.
