I have been having this issue with my 2000 c5. I programmed the Fob to the car and it works great till recently. Everytime I go to use it, it wont work till I hold the lock and unlock button down simultaniusly till the horn chirps. But less than 20 min later, you will have to do the same thing again in order to use the fob.
The FOB is bad. Trust me, you can try new batteries and reprogramming, but the 2000's seem to just take a ****. Dont buy a used one, get a new one for 2000 only.
the battery in the fob may not be making good contact. try taking it apart and cleaning the contacts on the fob. (i guess while you are waiting for the new fob to arrive )
If anyone ever has a FOB that needs to be SYNCED after starting the engine, that could possibly indicate that the 12 VDC main Battery is weak and may need to either be recharged or even replaced.
Don't waste your time, buy a fob. If you search here you'll find dozens of threads about the 2000's doing exactly what you describe. Gene Culley advertised them for $59 last time I checked. Buy the 2000 fob and be done with it.
Yeah, the fobs appear to have a lifetime. After 8-10 years, they seem to die for unknown reasons. I've contacted GM directly about this and all I can get from them is to contact a dealer. I know the new ones are assembled in Mexico but can't find out which company nor can I get a schematic for them. Not sure what all the C-5 owners are going to do when GM stops having them made.

I know...but I was thinking of the alarm functions. I think it will still work if the door is locked with the button before closing the door, then having x number of seconds to start the car once entering it again.
Worse will be for the C-6 owners if their fobs end up having a lifetime as well, since there essentially is no key. Fob doesn't work, car won't run on those.

The alarm will set and disarm with the key, just like the remote. If you unlock the car with the key, you don't have to start the car within any time frame, the alarm is turned off. Only if you reach in and manually unlock the door will the alarm sound. (Of course, window would have to be down (or broken out) or the top down to do that.
