My 98 has no differential gear whine on acceleration or deceleration but at about 70 mph under steady load there is a noticeable and annoying whine.
Is there a way to:
1. Measure backlash with the differential installed in the car
2. Adjust the backlash with the differential installed in the car
I can't find anything in the Helm manual on the topic but that doesn't mean anything. The twits that put this manual together have composed the most confusing, convoluted, practically impossible to use index on the planet. And to their credit, the same degree of obfuscation carried over from the C4 Helm manuals as well.

First I would make sure you have a diff whine rather than hub bearing/trans noise. If you do have noise from the diff, the ring and pinion gears are already on it's way out. Only way to check the backlash is with the diff out (you can access the ring gear from that end) out of the car unfortunately. The correct backlash at set up is 0.008". Good luck.
C5/C6 differential is a LOT different than any thing that your use to dealing with. Its unique and interesting. I recently up-graded mine to C6 ZO6 heavy duty parts using these post.
Like Rick stated, if yours is actually making noise, it needs a lot more than a back lash adjustment.
I purchased the Kent Moore rear end tool so that I could one day set up my own rear ends. Its a LOT more work than just using the tool. You need all the proper shims and a press and lots of time.
I felt really good doing the C6 ZO6 parts up grade!!
