Pull the plug and ground it to the block and see if you are actually getting spark whrn the engine is being cranked

Misfire can also be caused by the following:

1. Low Compression
2. Bad/plugged Injector
3. Bad driver circuit in PCM

Start simple

Do you have proper compression?
Is fuel getting to the cylinder?
Is that plug getting SPARK?

Bill thanks for the reply.
I will say that it runs great and you can go out for a ride and it will be fine then when you shut it off and go back to it with in say a 1/2 hour it will miss one time and set the code and then runs fine.
I will check the things you are saying.
Thanks again

try moving the plug, wire, coil, injector to another cyl and see if your misfire follows (one at a time would help determine what item).. if so you have narrowed it down to one of thoes items. Inspect the wiring harness for the coil on that cyl and make sure it has not been pinched/broken or scraped. does the engine make any abnormal noises? might want to pull the valve cover and look for a broken spring, bent pushrod and damaged rocker. After that a Comp test will tell you if its a mechanical issue rather then a electrical. rings, chipped piston, bent valve, ect.. Hope for the best. If its not consistant i would think it wouldnt be a mechanical issue.. but easy enough to check.

Look very closely at the things that you CAN NOT swap! Use a magnifying glass and look at the coil plug connector female pins. Make sure that they are not bent and or deformed. If they are spread apart, the coil will not have proper connection. Go to the auto parts store and find a new male pin the same size as the male pin in the coil connector. Insert it into the coil connector female pins and make sure that the female pins have good grip on the male pin. Should require some force going in and pulling out.

Remove the wiring harness covering on that wiring harness all the way back to the main harness at the back of the engine. Find the wires specifically for that coil and make sure that they are not damaged. Look at the point where the wires are crimped on to the pins on the coil connector.

You are running out of easy options. If all that looks OK,, you most likely have a coil driver in the PCM thats weak or going bad or the connections/pins at the PCM could be suspect

Common power and ground are good because all the other coils are operating SAT
