Rear End?

Rear End?
Good evening all,
I started my 12 GS A6 and let it go through my normal warm up process. Shifted into "D" and did not feel the transmission kick in, just figured I had shifted to neutral, so without looking I shifted into "S" and still no movement and no pressure on the brake pedal.
At this point I tapped the throttle and heard and felt a HUGE clunk from the rear end. I have taken the car out 2 or 3 more times since and no issue. Any clues??
I will add, that I have a 2x8 in my garage, so when I hear my rear tires touch it I know to stop (the 2x8 actually slides) but when I do touch it I always put it in gear and pull forward a bit as to not leave any stress on the transmission or rear end.
