Driving my (new to me) 2010 GS home for the first time the Service TPMS came on my DIC
I checked the air pressure with a gauge and the left rear was a little low at 26psi. I fill them all to 32 PSI and the warning went away.
Coming home today the alert came on again. This time I checked and the pressure was 32 PSI. When I restarted the car the alert went away again.
I am running Michelin Pilot Sports and not the OEM run flats. Could this be the problem or is it time to buy a new TPM?

Sounds like you mounted the Michelin's when you bought the GS. If so, did you replace the wheel sensors at the same time? If not, the original sensors are 13 years old & way past their battery life, which will cause
the odd readings. Whether you run RF's or non-RF's makes no difference. Again, if they're the original sensors, they need to be replaced.
I agree with above. If these are still the original sensors they are likely the issue and fading out. If you lose a sensor all together, you'll likely see "xx" on the display meaning it's not communicating. You can get AC Delco replacements through Rock Auto or even Amazon for a good price. The type of tire would have nothing to do with the sensor function.

The Michelin Pilot Sports were on the car when I bought it. The first time I got the alert the rr sensor displayed XX. But then it was fine and the last time I got the alert it read the correct pressure of 32 PSI. But, I am pretty sure that they are original to the car.

Based upon your update, I would have to assume that the sensors are original & do need to be replaced. As others have noted, OEM replacements (Shrader) are available from several vendors or whatever tire shop you will be dealing with may have them. The shop price may be higher but they may give you a break on the cost of replacing & programming the new sensors.
