I just got a new Z06. Can I effectively tune the car with a handheld tuner or will I need to find a tuner with HP Tuners in order to get it right? I do plan to add headers and other mods to the car.

I'm assuming that he messes with the LT1 and LT4 engines. He definitely works magic on LS3, LS9, and LSA engines so I would think that the works on the Corvettes too. The guy knows his stuff for sure...
Make sure whoever you go to TRULY knows how to tune Gen 5 / DI engines. It is a different ballgame. Find someone that a lot of customers have used and had their cars hold up, especially if heavily modded. There are some things that comes with tuning them that were not present on earlier models. A lot of tuners have no idea how to deal with DI.

I will go even more here and say 8 out of 10 that think they can tune will hurt your car are make it even worse with the tune they put into your car. Robert
You could talk to chuck "cow" with corvettes of Winchester that does remote tunes using autocal. That is they way I'm going with my A&A install as soon as I can get 3 days off in a row.
Just on the off chance that you aren't aware a tune will void your GM warranty. Not telling you to hold off but it is something to be aware of, it may not be worth the extra power if something goes wrong in the future.
