Yes, the vent tube will be used, there will be a vent port on either side of the battery, & your new battery will come with a plug for the side that you do not use.
Batteries are not sealed - they would pressurize as weather and altitude change. I sure would not want a pressurized box of sulfuric acid in my trunk. They are really just (owner) maintenance free.

So - some kind of pressure relief valve is present in a "sealed" battery. Should be safe enough...
If the battery has vent ports, plug one and run the vent tube to the other port.
