Having driven both cars in previous year versions 2007 IS 250 and 2008 G35 I would say the Infiniti hands down as it was much quicker and responsive along with having more interior space..... the Lexus has a slightly better build quality but its not enough to sway me. Now if it came down to the IS 350 vs. the Infiniti that would be a tough choice.

I've driven both and I like them both. I give the nod to the Lexus b/c it has better styling, fit and finish, softer leather, and toyota reliability. The G has more power so you should be comparing the IS350. It would be a landslide victory then.

We compared the G35 and the IS350. We ultimately chose the G due to extra space(needed for child seat), $ savings, and the ability to get the Sport package on the G.
She sure did love the IS350 though.

Do some research, Toyota has had a lot of problems of late (and I am NOT talking about the stupid floor-mat thing). My '06 IS250 got bought back under lemon law, which is very common for '06 to mid-'07s.
Since the OP is looking at '09s though the issue I had (carbon build-up/idle) should be resolved. However, the IS250 is a pretty slow car as compared to the G37. I like the look of the IS better, it actually handled and stopped very well (I even AutoXed mine a few times). It is pretty darn cramped in the back though. If MPG is a consideration mine got ~33mpg on the freeway before the carbon issue got bad.
As everybody has said though, the IS350 is what you should be comparing to a G37. I'd take the IS in that pairing even considering I had an IS that was a lemon!

I would pick the IS350 over the G37. As others have mentioned, the IS250 is not in the same league, powerwise. I have heard that a new M37 is coming out. If so, its price may be close to that of the IS350.

I have an 2007 IS250 and a 2004 G35 coupe. They seem pretty comparable in performance but the G would probably pull a little on the IS. The IS has more gears to it so it picks up pretty quick. The Lexus is tighter in quality however we bought it new and my G was used. Handling is about the same however a different feel. The only negative factor in my IS (or my previous GS430) is that they are dead quiet. No exhaust sound at all.. whereas the G sounds absolutely great when you open 'er up.
Overall, for a 4 door sedan, I'd have to go with the IS but I would get the IS350 or ISF . But for a coupe, the G37 hands down. (I'd go for a G37 over a C5 coupe too... but I'd catch hell for saying that in here)

I actually just bought a G37 S coupe and was cross shopping the IS350 and 335i. I would of personally gone for the 335, but couldn't find one anywhere close.
I really don't think you could go wrong with either, but most would say the G is overall winner. Both can have backup cameras and nav, IS can have cooled seats which may be a factor.
Not sure about the sedan, but my coupe is pretty firm - almost too firm for a daily over crappy Missouri roads.
