Does anyone have any idea on what the guidelines that Florida uses in putting a tax on vehicles on initial registration? I have heard that they randomly select and if it is too low they will ask for sale price verification. I was thinking that a 2000 model was worth about $12000 which would be $720 at 6%.

Get a bill of sale for the vehicle and bring it with you. If the price seems too low to them, they get out the NADA Price Guide for reference. I've never had a problem with them. As DMV's go, they're pretty good.
Yes, they will audit you...when I bought my vette I got it at a great price (needed work)....a month after I registered it I got a "love note" from the state revenue department...it said they thought the car was worth more and wanted to charge me the difference plus penalties and interest totalling about $1400.00 more..yikes...
I had to send a notarized affadavit explaining the purchase price AND so did the seller...real pita...it went away but chasing down the seller and getting him to do his part was no fun...
p.s. the state did not care about my bill of sale...
