It really depends how complete you want the paint job to be.
I took mine out to do the edges of the doors and the front surround, and then put just the doors in place (without the headlights) to paint the car. I took the doors back out to put the headlights back together and put the whole works back in. A LOT OF WORK!
So... how thorough and detailed do you want the paint job to be? What are you planning to do with the rest of the jambs, edges, surround gutters, etc?

well I will be removing the doors so a can paint the jambs, I would like a nice paint job with out going to crazy. would be a body on rest stow.
Frankly, except for the high dollar show car, I would paint at least the final coats with the car assembled. Aligning doors, headlights etc. is tough enough without worring about screwing up new paint.
