If you remove the 2 upper track 'stops' you MAY be able to wind the window high enough out of the door to replace what ever part has come loose.
Be sure to properly support the glass as it raises it's apex.

If you are referring to the roller that is in the front vertical window track.
Then chances are the parts are still in the door and you can get them to go back on.
I am not there to see what you have...but to answer your questions.
NO...the entire door does not need to be taken apart.
NO...the latch mechanism does not need to be touched.
NO...the regulator does not need to come out either.
YES...the black panel in Alan's photo (at the bottom of the door in the center) needs to be removed.
And YES...if you can not attach the parts that fell off and need to remove the door glass....let us know so you can get the procedure....if you can not find it somewhere the internet of forum. I have done more door glass replacements and removal/installations that I have lost count.
I hope you get this...because it is going to be really confined in there when you go to try to get it all tightened up....and hopefully the front roller does not want to spin when you go to tighten it.
I can tell you this. If you grab onto the threaded shaft of the front roller...and you can move it in and out and that 'in and out' play is IN the roller wheel itself. It is time to replace it....or you will have nothing but problems in your door glass due to excessive movement of the glass that is not supposed to be there.
The same goes for the rear roller wheel that is in the rear vertical track and also attaches to the horizontal guide for the door glass.
sounds like it is time to take the glass out and clean & lube all the parts! on both sides to forgo future problems.
The glass does not need to come out to do this. I lube up window assemblies quite often and I do not remove the glass to do this.
If I start to take parts out for a repair....that is when I inspect the parts like a hawk and replace them at that time. Pointless to reinstall worn out pats that I have lubricated.
