My butt hurts!
After a 100+ mile trip, my seat starts to feel uncomfortable. Other that replacing the seat padding has anyone done something different to make the seats a little more comfortable?

I do not think I will do it on my C5..........but on my C4 I always keep a set of plush lambskin seat covers on the seats(I do a lot of long distance driving). Those covers are great in summer and winter plus they are so soft. Everytime I get in the C5 I miss them..............just a thought...........

That's weird, I drive 300 miles every weekend from Hermosa Beach to Palm Desert in my '04 CE and never have any problems with the seats being uncomfortable.
Maybe your seat foam has started to disintegrate? Perhaps check into replacement foam? Either that or try this:
But seriously, good luck and hope you get this sorted out!
I drove from IL to CO... about 1100 miles... Awesome Drive!!! No Pain
Coming back to IL from CO was different!! I was hunched over for the whole trip because there was ICE/SNOW all over the Interstate!!! I went 55-65 MPH the whole way... After almost slding off the road 3 times I slowed down more and more!!! So the HUNCHING was not a good idea!!! But I still averaged 28 MPG!!!
My wife says her side (passenger) is uncomfortible after so many miles. My seat is fine, never had a new one so mot sure what a new seat would feel like. I would like to know if new seat foam would work.
Had an 01 Coupe a few years ago and drove it to Vegas. It tended to get a little uncomfortable after a long day of driving. The side bolster on the bottom part of the seat seamed to have began to wear and didn't provide support it should have. Have an 02 now and the seats had some extra padding put in them when they were redone and I have noticed they are quite comfortable. Took a 1200 mile trip a month ago and had no problems. The extra padding keeps the leather taught improving the appearance of the seat also. Very happy with it.

According to your profile, you have a 98. If you haven't had the foam replaced, you are way past due. The early C5 foam starts to break down after just about 5-6 years. I changed mine this spring on my 99 and was amazed at the difference.
When the C5 was introduced, there was much kerfuffle about the seats. They were definitely a step back from the C4, which had seats designed and supplied by an outside firm that specialized in seats for high performance cars (the name slips me right now).
I found that to be true. My old C4 seats and steering are the two things I would transplant to the C5; the only things I would transplant from the C4.
That said, it's all about how one is built. The C4 was great for comparatively thin and average height people, I'm 5'10" and 165. That is why they changed the seats in the C5: we are much heavier and getting taller as a people.
So for some folks, the C5 is more comfortable, and for others, the C4 is preferable.
I also do not find the C5 seats to be comfortable over long hauls. But everything else makes me forget about it, until I stop for the night!

Upon first look I thought you were taking a very long way around to get from HB to PD. Then I realized you probably were counting Round Trip. Thank god it is the weekend my brain turned off around 2:30PM and is running on the reserve tank.
