At this point, Id say you could squeeze a few more ponies out with some custom PCM programming, or using a MAFT with Autotap. Mabey not worth the trouble and money. Id save for a cam or cam/heads. Or mabey even a 3.73:1 rearend.
A cam will get you 25 to 30 plus HP. for under a grand.
I agree with Kewlbrz. You could probably do about 90% of what custom tuning will get you with a MAFT and using someone's autotap. I have a feeling all that would need to be done is have your car richened up a bit to make up for the extra air you have entering your engine.
A heads/cam package or cam only would be my next step down the mod path if I was you.
I've heard the waiting list for Ed is up to 6 months. If I were you I would get on the waiting list around 5 months before you plan to get the heads/cam. After you get it all done you should give me a ride :D

No that's not correct either. Since Steve's recovered from the mnay surgeries due to the motorcycle accident last year, the lead time isn't 6 months. Don't think Ed Wright's is either.
I've had my mods for almost a year, but my car has never been tuned. Would it do me much good at this point to get it tuned?

I went there last month, & put up a post about my results. I gained 20 RWHP & 20 RWTQ across the RPM band with his tuning. The results are in my sig. He's VERY good. I drove round trip from Houston. You're only an hour away. I highly recommend his services.
I’m in the same situation and I decided to save my $$$ and wait until after I do heads/cam & gears. Then I’ll do a dyno tune with Ed. IMO the performance gain on a stock motor is not worth the $$$.
I noticed that you live in OKC and have almost exactly the same mods as me. Let me know if you would like to hook up and run our vettes on the street and/or highway. I’d be curious to see how my LT4 would do against a similarly equipped 2001 LS1. If you’re interested my mods are:
K&N Air Filer w/ open air lid
NGK plugs & MSD Wires
160 thermostat
Flowmaster cat-back exhaust
DRM long tube headers (no cats)
Stock everything else (including tires) :sad:
