You'll lose some coolant but not near all of it. Personally I wouldn't want to try and flush a system just after new head gaskets and or heads. I personally would, if my coolant was pretty old simply drain the rest of the system down, by removing the radiator petcock or lower radiator hose, to remove as much coolant as possible (good idea anytime you will be removing engine parts with coolant in them anyway before you start, to keep coolant from getting places you don''t want it). If when I had the engine apart, and saw the need for a flush (scale, and other deposits), I'd wait till I had some time in with the new heads to make sure everything was good and proper there, and then flush it.
You will loose 3/4 for the coolant. The rest stays in the block.
I just use a wet and dry vac and suck the block out. MAKE SURE that you get ALL the coolant out of the block head bolt threaded holes or you will destroy the block when you torque the head bolts on.
I used an air chuck with a long thin metal tube to blow out the holes
