Been reading up and seem to have this narrowed down to two possible culprits.
Problem: Driver's door locks fine but occasionally will NOT unlock with the door button or the key fob. You can hear something clicking near the speaker but the door won't unlock.
When that happens:
Door locks fine with button, key in door, and fob.
Door only unlocks with key in door
Question: Should I be looking at the actuator or one of the three relays?

There is only one actuator that handles both locking and unlocking motion. So, barring an unusual mechanical linkage problem, it's fairly safe to conclude that it's a circuit problem. A burned or dirty relay contact would be my guess but it is only a guess. The way to find out is to get in there with circuit diagram in-hand and eliminate the possibilities with your multimeter.
(The two relays that energize the actuator are normally grounded. When a relay is activated it changes from ground to +12V. So you need to first make sure they always provide good ground contact and that they provide full power when energized.)
Let us know what you find.
