Default Push rod length / lifter preload

I've read and watched a number of How-to's regarding checking push rod length to get the right preload. My question is does it matter that I'm installing new lifters? (LS7's). Since they haven't been pressurized yet, isn't that going to affect my measurements?

No, it won't affect the measurement itself however it will make it more difficult because the plunger will be very easy to depress while you are measuring with the adjustable pushrod.
