I don't know about other dyno's but on the dynopack that my guy uses he changes a setting to compensate for the ratio so that you get a correct reading.
By the way, there is not a 1 to 1 gear ratio in the A6. Run it in 4th, it's the closest at 1.15 to 1. If you run it in third, that's fine, you'll just show a little less horsepower. And, of course, this is perfectly fine if you are using the dyno for what it's meant to be used for, a tuning tool. Just make sure you dyno it in third again after you do your mods and/or tuning. A dyno is not that good for an absolute number, but it's great for a comparison.

I do them in 4th gear here, as they said there is no true 1:1 like with the manual cars.
Unlike the older 4spd cars that had to have a Tech II or laptop to keep the car in gear, the paddle cars will stay in gear during sport mode which makes is very nice. Generally do a 2500-redline pull depending on the cam setup what redline would be.
